It's every two minutes. So if you pick up the rune at 1:59 the next will spawn right at two minutes.

iloveroo, notice that if there is a rune still there (Top or Bottom) on the 2 minute mark; a new rune won't appear. Not two runes at once, not replacing old rune. It will just not happen.

Lets be clear bout the runes.
Every time the number of minutes on timer is a multiple of 2(starting at 0) it checks if a rune is on the map. If there isn't, one is spawned at random ,top or bottom. It is absolutely possible to have haste and dd at the same time if you're super lucky and had good timing .
I remember once when I played a ganker with bottle I would pick up DD and it would still be there, on the ground and in my bottle. I was like WTF is this a bug, but apparently i picked it up exactly the moment a new one would spawn :D

since there are so many leavers right now, what are some tips you would give when playing a man (or men) down?
i know the usual, play conservatively, farm as much as you can while you can, don't overextend... for example, if i'm windrunner, do i just try carry instead of support? do i try gank more?

You need to outfarm the enemies or win by push. Best way to do that is to go all 4 and gank them quickly when they don't expect it(depends on phase of the game, the earlier the leaver is gone, the more your gank of 4 will be able to achieve) and than push REALLY HARD. If the game is in early/mid stages playing with rax down is really hard. Ofc such a push is a risk but you need to risk when you play at disadvantage, right?
It is key to not let the enemy capitalize on their numbers, gank 1 or 2 people, stick together(but not to close if they are aoe heavy xD) and earn as much gold as possible if you can't win by push, so you can make up in items what they have in numbers.

About the autobanned heroes in -cm (these should be terrorblade, roof, pitlord, rubick, thrall, io, tuskar, phoenix). What is the chance that these heroes will get available for competitive play soon? Thrall and Rubick would probably change the metagame/be instabans, but otherwise?

I'll answer the unasked question, they're not avaible in cm becuase they're not balanced because they're new heroes or were recently remade.

Phoenix would be instantly banned every single game along with the other ones. His ulti is just too good in combo with the other skills. He is probably the best team fight hero in the game at the current moment.
Basically speaking the meta game would change if any one of those heroes were added since they do things better than other heroes already in the game.

Thrall's spells also combo greatly with most initiators, his ult and movemnet blocking spell is really deadly. I remember it was enough for me to spam all skills and die to get triple kills the one game I played him, his aoe potential is INSANE

Is there any significant differences in gameplay and design between DOTA 2, Blizzard DOTA, HoN and LOL? or are these games all essentially the same game, produced by competing companies/organizations?

There are significant differences. Each one is a different game, from the same genre. Is a little like trying to compare different Shooters or RTS. They have the same basics and idea, but they play in different noticeable ways.
The exception could be HoN, which was original a port (copy-paste) of DotA, like Dota 2 is doing now. However HoN chosed a different path some time ago, introducing some minor gameplay changes and new heroes (and I believe items aswel?) that are not present on DotA nor Dota 2.

depends on the hero.
as enigma, your starting items could feasibly be 12 mana potions (buy tp & bootsfrom sideshop later) then basilius, soul ring, treads, blink dagger
