A patient enters the hospital with an invasive tumor of the midface involving largely skin, cartilage, and bone. The tumor is resected by the head & neck surgeon with clear margins. The patient received postoperative irradiation to ameliorate any residing metastases. The patient possesses a large midface defect. As a tissue engineer, you are working with the reconstructive surgeons to create a construct that will fill the defect. You think that a new form of hydroxyapatite (HA) that you have developed will permit the formation of a three dimensional construct that will integrate well with the patient’s anatomy. The new form of HA is functionalized with osteoblast (OB, bone forming cell) adhesion peptides. The material is denoted HA-OB. An obvious design question is whether HA-OB possesses the mechanical strength required to support the midface. You decide to conduct a uniaxial tensile test of a 2 cm diameter, 3 cm long cylindrical core of HA-OB using an Instron. A 100 kg mass is used as the load. The HA-OB elongates 30%. (a) What is the Young’s modulus for the HA-OB? (b) Based on the Young's modulus you decide that HA-OB meets your mechanical design constraints. A 3D construct is fabricated and is ready for the operating room. What major constraint may limit survival and integration of the construct (which is seeded with osteoblasts) with the patient's anatomy? Explain why. (c) In another application related to bone repair, poly(lactic acid) (PLA) is being investigated for its use in bone pins, as a multifunctional material that helps correct bone defects and fractures. Its two main functions are to provide mechanical support and to release bone growth factors to stimulate growth. What property of PLA can be readily modified to give it the appropriate mechanical strength? Explain why. (d) For this particular application, what relative level of crystallinity (high semicrystalline; low = amorphous) would enable PLA to effectively release the bone growth factors over time? Why? (e) Keeping your answer above in mind, describe the properties of PLA that contribute to its crystallinity and its ability to deliver growth factors in the desired way. What processing conditions could help reinforce these properties in PLA? How do these help? | ||

Today, I arose from my peaceful slumber to the sound of teeth chewing through my various plants. I pushed the marble statue of myself down the stairs, so the beasts would think I'd fallen down, and hopefully would continue munching, blissfully unaware that I was climbing onto the roof with my electronic crossbow. (its cool, it has a cell phone and mp3 player). Yet when I got to the roof, they were nowhere to be seen, like jewish people at a "manual labor" festival. My question for you, Sir, is how do I rid myself and my bountiful garden of these creatures? | ||
