Sheng villagers ("peon") were 30% cheaper than every other civ's workers in Age of Empires 1. No drawbacks. No penalties. Just incredibly broken cheaper workers.
Imagine if SCVs cost 35 minerals instead of 50...

Street Fighter 2, original arcade release: Pick Guile, camp in the corner charging your flash kick. FFVII: There's a mid-game weapon for Vincent called SniperCR. The damage is only so-so but its gimmick is that it has 255% accuracy. Reach into your inventory and pull out that Deathblow materia (double damage, half accuracy) that has been useless until now. The next several hours are easy.
Super Mario World: In Mario 3 it was unclear whether you wanted the raccoon tail or fire, since the raccoon tail's attack is short range and hovering requires mashing the button in a kind of uncomfortable position. In SMW, holding the button lets you hover with the cape, the attack range is improved and it doesn't delay your run, and you can still use cape+Yoshi. It's clear that you want the cape in all circumstances except for maybe 1 of the underwater levels. Note that in the DS NSMB they finally realized that mobility powerups were all the best in the Mario games and nerfed the mini-mushroom so that taking it makes you risk dying in 1 hit.
Morrowind: First thing you do is grab a decent mortal and pestle and start practicing alchemy. Alchemy lets you make potions to buff your stats, including those stats that govern alchemy. Ride the positive feedback loop for just a few minutes and you'll have stats that exceed the naturally achievable limits by a couple orders of magnitude. In theory these potions will wear off sometime, but by the time you've boosted your stats that high they'll last for hours at a time.
oblivion, master the mage guild and create two spells that buffs your mana/regen and provides -magic resist keep using both one after the other a while then switch to the most powerful other spell that you could never have cast otherwise (requiring say 20000 mana) and one hit kop whatever gets in your way

pistol from Halo:CE 3 well placed shots could kill someone soooo quickly. one clip could potentially take out 4 people in about 4-5 seconds. good range, and very accesible (often you started with one)

World of Warcraft pvp season 5: Yeah I'm not expecting this game to be completely balanced, there are so many classes/specs/situations.. but this point in time was just pure comedy. DKs aoe stuns, self rez, and 15 pets, ret paladins occasionally 1 shotting people, paladin talents being completely screwed allowing for insane healing with half their points in a damage tree, infinte mana, etc. Fun stuff.
Empire Earth II - that Chinese fire archer special unit that they got from what I think was just the first building you could build combat units from. Except that it completely owned every other unit that anyone else could get out at the time. So you could just funnel fire archers into the enemy base and they would keel over.

I really enjoyed the time when druid trees couldn't be killed in 2s. It was impossible to mana drain them and kill them.
