Does this work for replays from older versions of the game? This is a new feature coming with the update that will ship with Heart of the Swarm. Unfortunately, previous replays from older versions of Wings of Liberty are not compatible with this feature. Only new replays created after we release the update will be usable with these features. At that time, both Wings of Liberty and Heart of the Swarm players will be able to use these features on new replays.
Does this work with custom maps?
These features were designed with StarCraft II melee maps in mind. Due to the variety of trigger functions usable in Arcade maps, we are currently not officially supporting these features in Arcade maps. There may be issues when using these replay features in Arcade maps. Having said that, there are plenty of Arcade maps that will work with these features, so we are not disabling the functionality at this time. We hope to spend time in a future update improving the Arcade support of these replay features so that it will work correctly in all Arcade maps.
If you load from the game, does it create a new replay file of the old part of the game + the new part of the game -- and can you then load from THAT replay?
Yes! If you take command of a replay at some point while watching it and then finish the game out, a new replay is created that has the original first part and the rest of the game as you played it out. You can technically keep doing this with these new replays to create all kinds of interesting situations! The point at which the players used the Take Command feature in the replay will be marked in the new replay with an in-game text message that indicates that new players have taken control of the game.

For the Take Command feature, can you choose an AI as your opponent?

Really liking the info, here. Resume from replay alone will be so amazing for tournaments. Now it won't matter if the internet drops or if someone blue screens, there should never be a need for a regame again, except in the case of total venue power outage

Someone else asked that, so far I havent seen blizzard say one way or another (although my guess is you can?).

You may even be able to in the case of a "total venue power outage" if you got an uninterruptible power supply on one of the computers (like the casters computer). You just need one person to have power for a minute to save the replay (and I dont think you even need to be connected to battle.net for that).

Here's a question: will this resume from replay feature ONLY be available in HotS? (Probably yes?) And if so, if I own HotS, will I be able to reload WoL games and resume from replay, too?
If marketed correctly, I think this future could give a noticeable boost to sales of SC2/HotS. Right now there's a certain segment of the SC2 viewer audience that ONLY watches and doesn't play the game. But if you could tell them, "Hey, now you can jump in the middle of that Squirtle vs MVP Game 6 GSL Finals battle versus a friend", they suddenly have an incentive to learn how to play the game and buy a copy.
Speaking of which...... I'd like to try Game 6 and Game 7 from that series

Not super practical for any sort of open bracket play, but for stage matches you are 100% right

I dont think you can use WoL to load HotS replays or use HotS to load WoL replays....

The "Take command" thing is really really cool. Makes me wonder if we can "take command" of pro replays, or if its player specific.

Thank you Blizzard, it is about time friends could once again enjoy replays together ...

Both WoL and HotS will use the same client once HotS comes out, so you'll be able to load replays from both games within a single client.

I believe they have stated you won't be able to resume the game from an old replay. You'll only be able to do it from a replay generated after this feature has gone live.
