After watching the new wrath movie, I came to an unsettling realization... Diablo the big badass boss of the Diablo franchise may not even be in the game! We may never encounter him as a boss.
It is possible that the scary Diablo seen on the box and in the cinematics is the representation of the 'true' form of Diablo BEFORE he was overthrown into Sanctuary. The cinematics of Diablo could just be a flashback to explain the lore, like a deckard cain story. This is why i think Diablo the Lord of Terror may not even come up as an act boss in the game:
1. Diablo was slain in D2 by the hero, how could he magically obtain such a powerful form after only twenty years? Remember that after the prime evils were cast out, Diablo was in a weakened state such that he couldn't even posses King Leroic. He had to gradually gain power by swapping bodies. Unless the breaking of the soulstones somehow unbound his powers so he could come out in his ultimate form. If this is the case, then why did Tyreal tell Marius to destroy the soul stones at hellforge? It seems odd to me that Tyreal didnt tell the hero in D2 at Pandemonium Fortress to trap Diablo instead of slaying him, since Diablo is an entity that is not destroyed when the phyiscal vessel is destroyed (as seen Wrath).
2. Could he be an act boss? Well we already know that + Show Spoiler + is most likely going to be the act 1 boss. I think it would be highly probable that Azmodan or belial be an act boss in either act 3 or 4. Other possible bosses that come into mind in acts 2-3 may appear: mistress of pain, zoltun kulle, imperius. Does that leave any room for Diablo to make an appearance?
I for one would be disappointed if I did not face Diablo in battle in Diablo 3. But after what i have seen I am fearing it might be true. What do you think?

+ Show Spoiler +I think it's pretty obvious after playing the beta, that it has to do sth. with the meteor crashing into the citadel that he will get resurected (I mean it resurected leoric so why shouldn't it has the power to bring Diablo back

Blizzard has already answered this:
+ Show Spoiler +Diablo will be the boss of Act 4.
Apparently, after 1.5 years, this is my 100th post.

The soulstones were a sham according to Izual, the girl leia is the daughter of the D1 witch and the hero who defeated Diablo, later becoming diablo in Diablo 2. So just here there are two possibilities of how he returns, three if you add in the meteor, four if you consider the black soulstone. He's in the game allright.

Also people seem to forget that there can be more then one boss per act? Diablo is obviously a main boss but for characters like Imperius, Mistress of pain and whatever an Izual:esque appearance is perfectly fine.

+ Show Spoiler +It's not official. Based on profile/achievement pictures they found. The boss of each of the acts has been heavily speculated, even the final boss is open for debate. That could all now be settled with the discovery of another image post in the Beta Forum, apparently boss profiles that will appear in your profile once you have defeated each. Each has a picture and the Act number above them. from: http://diablo.incgamers.com/blog/comments/act-bosses-confirmed+ Show Spoiler +imgur not working for me at the moment, sorry for wrong scaled pics... Imperius is speculated for an expansion boss.

+ Show Spoiler +Imperius and Tyrael never got along very well. Imperius also afaik doesn't share Tyraels concern of humanity. Also if you check the recent animated short film released, and listen to what Diablo says and how Imperius acts throughout the movie you'll get a little insight to how he is as a "person". He will stop at nothing in defeating the burning hells, it is all that matters. He'd gladly sacrifice legions of angels and the entire human race to accomplish his goal.

I'm the second no Diablo vote. This game is going to be all about corruption and the lesser evils taking control of hell's forces. Diablo will make at most a cameo, nothing more.

From the wiki about imperius: + Show Spoiler +When Sanctuary was discovered by the Angiris Council, he was the first one to raise a voice against its existence. He regarded humans as abominations polluted by the presence of Demon blood in their veins. He tried his best to have the hidden world destroyed but was opposed by his counterpart Auriel, eventually resulting in a Council vote to decide Sanctuary's fate. The vote went against his favor when Tyrael shockingly voted for humanity, but Imperius still harbors a deep grudge against the "demon-spawn" humans.

I will only be satisfied if Diablo gets to be the last boss of the last expansion.

If you've watched the Wrath movie you must have seen the part where Tyrael says that if Diablo is killed instead of imprisoned, he will COME BACK.
To me, that means he's a boss.
Also, you can't have a Diablo game without yours truly.
