So, I just played through Diablo 3 for the first time. I played a barb start to finish just going through the main story and got to 31 after defeating Diablo. I know there is a whole other game beyond the story that I want to experience but don't know where to start. So, given that, here are my questions.
1) What are my goals after my first play through? Do I essentially play through the game a second time on the next difficulty up, then a third and fourth time to reach and defeat the highest difficulty? Or, am I instead looking to accomplish some other goal such as reach a certain level, get a certain amount of money, or farm certain gear?
2) What should I be looking out for while playing? As a veteran of old school World of Warcraft, I know that I was KICKING myself about getting rid of any fire resist gear when getting close to the level cap (then ESPECIALLY nature resist and frost resist) because I didn't know that a big goal was to farm up this gear that allowed you to survive the raids I was going to be doing. Things I'm looking for here; materials I want to be farming for, items I want to save, Items I want to look at vendors for for a good buy.
3) How do I level up quickly? Weather I'm leveling up an alt or my main character, how do I Level up with the most efficiency?
I can't think of anything else right now, so there it is. Any help appreciated. Thanks!

For now, try to get the Hellfire ring in Inferno.

Hi
1) Play though again and again until you are lvl 60 and can start in Inferno. After that you need some gold to buy completely new gear because <60 gear does not compare to 60 gear.
2) No need to keep anything except if you should find Leoric's Signet. If you find good gear on your way to lvl 60 try to sell it in the AH. You can keep good gear you find for alts, but it is in no way necessary.
3) If you have a friend who wants to help, you can play on a higher Monster Power, so you get more levels while beating NM and Hell faster. Other than that just playing through NM and Hell normally should get you where you need to be level wise.

Thats all there is to diablo. Grind through the difficulties untill you beat the highest and are max level.
Then grind gear.

I mean, im posting it in a simple manner, but its the truth

Slap the most expensive red gems you can afford on your helm and weapons or get somebody to boost you to lv60 asap to reduce the tedium of repeated playthroughs. Nothing you find until inferno is even worth picking up (except the aforementioned Leoric's Signet and the occasional Homunculus).
The game begins at lv60. At lv 40-something you can start to scour the auction house for low-end lv60 gear with the affix Reduced Level Requirement.
edit: afaik, with a Hellfire Ring, Leoric's Signet, Cain's Set and a full party, you can go from lv1 to lv60 in around 2 hours with a good route.

Dont farm gear until you get to inferno, it's pretty much useless. So for now your priority should be to get atleast one character to inferno, make a character capable of beating inferno...and then farm for gear and other things.

Your pretty much done with the game. The rest is all the same but with bigger numbers.

Hah, as sad as it sounds it's pretty true. After killing Diablo on normal you pretty much finished the game from campaign point of view. Now it's time to do the same on higher difficulty and eventually get into the farm game, obtaining best gear possible, getting access to uber bosses etc. You can also try hardcore difficulty, which is way more fun (at least for me) since you need to start all over if you die. You can grab all non-grey and non-white gear and destroy it at blacksmith station to get crafting materials.

-Better/different gear lets you play around with other builds. The game is replayable with a different approach, granted the story remains but diablo is mostly about the gameplay. At level 60 you get to pick any talent or skill and you have a better variety of item affixes/suffixes.
-You have access to multiplayer if that's an attractive option.
-I'd advise against using the AH, defeats the purpose of clearing areas, altough it sometimes suck to beat on champions or bosses using an underleveled "spoon". I avoid shopkeepers and crafting (except gems) for the same reason. It helps in making the gear acquisition and zone clearing much more linear. To each their own.
