When you do Shift+1 it adds the selected unit(s) to the control group 1. But there is no way to remove them with a single hotkey?
What I usually end up doing is to select the group (hit 1), shift+click the unit to exclude, and then Ctrl+1 to reset the group.
For example I like sending out single zerglings to various locations for scouting. If they are already in my primary control group, which they usually are, I gotta remove them by going through those multiple steps, which gets tedious. Is there a better solution to this?

I'm not sure it exists. When I send zerglings to scout, I first give them the move order, and when they're away from the pack i just box-select my remaining units and reset the control group. If my remaining group is sparse and not box-selectable, I prefer to click on the zergling's icon in the bottom center part of the interface.

Have you tried selecting a unit already in that control group and then pressing Shift + 1 to undo (perhaps)?

um, no "easy" way to do it.... mostly just by selecting a single unit and reassigning, or removing that single unit and reassigning...

you can do that. try selecting the whole group then click SHIFT (or SELECT not sure) then click on the UNIT (you can click on the unit in the menu at the bottom) the unit will be removed from the selection. but you will still have to reset the group name. hope this helps

yeah as stated above thats pretty much the easiest way.

I feel really dumb for not knowing that Shift+1 adds the current group to the Hotkey group of units.
I always thought you had to have the hotkey group selected, press shift, select new units, and then Ctrl+1.

I usually order the entire group to move to the location Then shift+deselect a zergling via wireframe Ctrl+1 to rehotkey Move remainder of blob wherever

i woulda sworn it was someting like ctrl shift # or something like that.

I usually use ctrl shift click, shift click, or remake the ctrl group completely .

I have a question which fits in this topic i think 
If I have many overlords and i want to send them in different locations fast, how can i do that?
I remember Psy showed how to do that with shift, but i forgot that youtube link and when i try it does not work for me.

select a bunch of Ovies. right click location. shift click an overlord (either on screen or wireframes), then right click new location. repeat.
i do this, but even then it's not really that fast. it is still the fastest method and the only thing that can be done is improvement of hand coordination / speed with practice.

Shift-1 is very useful, but I kind of wish that shift+ctrl+# did what shift+# does now.
The reason is that I wish that shift+# would add the selected group to your current selection. So if I have marine/marauder in 1, vikings in 2 and tanks in 3 I could <hold shift> 1 2 3 <release shift> to select all 3 groups into 1 control group. I feel like I have to have a separate control group with all my units in it is unintuitive.

Totally didn't know about the Shift-1 feature. Totally not gonna affect my gameplay, it's not like I'm gonna go from 400 Diamond to 700 Diamond by this awesome technique.

I would kill for a "remove last unit from selected group" hotkey.
IE. You have 4 zerglings selected. You press the hotkey (say, ctrl+shift+F), and now you have 3 zerglings selected with the last one in the unit grid window having been removed from your selected group.

You want to move a zergling out to scout a location, but he is in your primary control group. You need to issue him a command then remove him from the group so that you can go on about your business. Try this: (pretty sure its the fastest way). Assuming control-group hotkey is 1
1) Press 1 2) Right click mini-map on the scout location to move out 3) Shift-Click the wireframe portrait of the zergling(s) you want to send, removing them from the selection 4) Quickly press stop/halt, which will freeze your remaining army 5) Press Control-1 to reassign the group, sans the zerglings you shift clicked 6) Watch those plucky little bastards run off into the darkness (to be killed horribly).
(pretty sure zerglings technically ARE bastards, so...)
