It actually doesn't because the creep tumor is placed so that you need to get rid of it before the Zerg can even make a hatch.

The Creep Tumor is situated in the location a hatchery fits on the island. There's no space to build a hatchery around the creep tumor and as you said, you can't build on tumors.
